All oligos synthesized at IDT are analyzed by electrospray ionization (ESI) mass spectrometry for quality control, ensuring that only the highest quality products are shipped.
Mass Spectrometry Analysis
CE provides quantitative analysis of micropurity. Nanoliter samples of oligonucleotides are electrokinetically injected into a fused silica capillary and filled with a denaturing gel under an electric potential. The oligonucleotides move through the gel matrix and are separated according to length. Absorbance at 260 nm is detected as the oligo passes through a window in the capillary. The resulting electropherogram provides the percent purity of the full-length product as well as truncated products.
HPLC is a universally accepted method of sample quality assessment. Both reverse-phase and ion-exchange methods are used at IDT to assess oligonucleotide product purity. These methods are used for purified oligos whose composition is incompatible with CE technology.
