What is the best storage condition for a custom gene?

Custom genes should be stored lyophilized at –20º C until the first day of use.

Prior to opening and resuspension, the gene tube should be briefly centrifuged to ensure that the lyophilized DNA is at the bottom of the tube.  Recommended resuspension method follows:

  1. Re-suspend lyophilized DNA in 20µl of TE buffer (10 mM Tris, 0.1 mM EDTA, pH 7.5–8.0) or water to achieve an approximate stock concentration of 0.1 µg/µl (100 ng/µl).
  2. Incubate at room temperature for 30 minutes, and then vortex for 20 seconds.
  3. Centrifuge the tube(s) at 10,000 x g for 1 minute.
  4. Create aliquots (optional).
  5. Store DNA at –20º C.
