Do I need to worry about performance if my shipment of PrimeTime® Gene Expression Master Mix is delayed?

No. IDT has tested 3 batches of PrimeTime master mix by heating at 50°C for 1, 3, or 7 days before use in qPCR experiments (Figure). High PCR efficiency was observed (range of 95.1–103.6% with R2 >0.991) with the assays tested (HPRT and GUSB). When comparing the controls (containing frozen, unheated master mix) to reactions containing heat-treated master mix, ΔCq values were <0.5, with a few exceptions that were primarily seen when using a low amount of starting template. The small ΔCq values emphasize the stability and robustness of PrimeTime master mix after extreme thermal stress treatment.

PrimeTime master mix was incubated at 50°C for 1, 3, 5, or 7 days, while a control master mix sample was stored frozen (–20°C) until use. The reactions remained at room temperature for 24 hr before running the thermal cycler: control master mix (gray) and the 1-, 3-, 5-, and 7-day, heat-treated master mix (green, light blue, dark blue, and orange, respectively).
