It provides information about organisms in areas as diverse as population genetics, epidemiology, organism identification, <a href=\"\/pages\/applications\/genotyping\"> genotyping<\/a>, <a href=\"\/pages\/products\/next-generation-sequencing\/archer-ngs-assay-solutions\/solid-tumor-research\/archer-variantplex-pan-solid-tumor-panel\">rare variant identification<\/a>, <a href=\"\/pages\/research-area\/cancer\/cancer-research\/early-cancer-screening\">cancer and rare disease&nbsp;research<\/a>, gene editing confirmation, and gene-environment interactions.<\/p>\n<p>NGS is a high-throughput technology that determines the sequence of a sample all at once by using parallel sequencing. Traditional Sanger sequencing determines the sequence of a sample one section at a time. Sequencing thousands of gene fragments simultaneously with NGS reduces time and cost associated with sequencing and increases the coverage quality and data output.<\/p>\n<h2>NGS workflow<\/h2>\n<p>Most <a href=\"\/pages\/technology\/next-generation-sequencing\/workflow\">NGS protocols<\/a>&nbsp;follow a similar workflow (Figure 1). The first step is nucleic acid extraction, either DNA or RNA. This nucleic acid material must be prepared for sequencing by converting it into libraries. <a href=\"\/pages\/education\/decoded\/article\/what-is-library-prep-in-sequencing\">Library preparation<\/a>&nbsp;involves adding adapters, which allow the samples to be indexed (barcoded and identified).<\/p>\n<p><figure class=\"wp-block-image\"><img decoding=\"async\" src=\"https:\/\/www.idtdna.com\/page\/wp-content\/uploads\/idt-images\/0bd47b15-3279-6e2e-aa53-ff00001c1b3c-22-ng-figures-xgen-ngs-oncology-research.png\" data-displaymode=\"Original\" alt=\"Typical NGS workflow\" title=\"22-ng-figures-xgen-ngs-oncology-research\" \/><figcaption class=\"image-caption\">Figure 1. Typical NGS workflow.<\/figcaption><\/figure>\n<\/p>\n<p> Illumina&reg; sequencing is performed using sequencing-by-synthesis on a flow cell. Short-read sequencing generates 100&ndash;300 base pair lengths called reads. The reads go through quality control and are aligned to a reference before being evaluated for significant characteristics.<\/p>\n<p> Learn more about NGS library prep and why it is important in the <a href=\"\/pages\/education\/decoded\/article\/why-library-prep-is-essential-to-sequencing\">Why library prep is essential to sequencing<\/a>&nbsp;DECODED.<\/p>\n<h2>What are the types of NGS?<\/h2>\n<p>There are many <a href=\"\/pages\/community\/blog\/post\/let-s-talk-about-sequencing\">types of NGS<\/a>&nbsp;for&nbsp;DNA, RNA, DNA-protein interactions, and <a href=\"\/pages\/products\/next-generation-sequencing\/methods\/methyl-seq\">methylation<\/a>.&nbsp;The different NGS approaches can be largely thought of as targeted and untargeted.<\/p>\n<p>Untargeted NGS, which examines all the nucleic acids in a sample include approaches like:<\/p>\n<ul>\n<li><a href=\"\/pages\/products\/next-generation-sequencing\/methods\/whole-genome-sequencing\"><strong>Whole genome sequencing<\/strong><\/a> <\/li>\n<li><a href=\"\/pages\/technology\/next-generation-sequencing\/dna-sequencing\/targeted-sequencing\/exome-sequencing\"><strong><\/strong><\/a><a href=\"\/pages\/applications\/infectious-disease\/shotgun-metagenomics-for-infectious-diseases\"><strong>Metagenomic sequencing<\/strong><\/a><\/li>\n<li><a href=\"\/pages\/products\/next-generation-sequencing\/methods\/whole-transcriptome-sequencing\"><strong>Whole transcriptome sequencing<\/strong><\/a><\/li>\n<\/ul>\n<p>While targeted NGS relies on primers or probes to target distinct genetic regions of a sample.
