{"id":1250,"date":"2020-10-05T16:36:46","date_gmt":"2020-10-05T16:36:46","guid":{"rendered":"https:\/\/www.idtdna.com\/page\/what-helps-supercentenarians-live-so-dang-long"},"modified":"2025-08-22T16:17:18","modified_gmt":"2025-08-22T16:17:18","slug":"what-helps-supercentenarians-live-so-dang-long","status":"publish","type":"post","link":"https:\/\/www.idtdna.com\/page\/support-and-education\/decoded-plus\/what-helps-supercentenarians-live-so-dang-long\/","title":{"rendered":"Why do supercentenarians live so long?"},"content":{"rendered":"<p>When we are talking about older people, we have octogenarians (people in their 80s), nonagenarians (people in their 90s), and centenarians (people in their 100s).&nbsp;<\/p>\n<h2>What is a Supercentarian and How Many are There?<\/h2>\n<p>And then there&rsquo;s a class all to themselves: supercentenarians&mdash;people who are 110 years or older.<\/p>\n<p>In all, some <a href=\"https:\/\/en.wikipedia.org\/wiki\/Supercentenarian#:~:text=Over%201%2C500%20supercentenarians%20have%20been,documentary%20support%20to%20be%20validated.\" target=\"_blank\"><strong>1,500 people<\/strong><\/a><strong>&nbsp;are supercentenarians. <\/strong>One in 1,000 people who reach the age of 100 live on to 110; several dozen are alive today, including a woman in Japan who was born on Jan. 2, 1903.<\/p>\n<h2>What do supercentenarians have in common to live so long?<\/h2>\n<p>Supercentenarians themselves cite a <a href=\"https:\/\/www.huffpost.com\/entry\/healthy-living-news_b_5301711?guccounter=1&amp;guce_referrer=aHR0cHM6Ly93d3cuZ29vZ2xlLmNvbS8&amp;guce_referrer_sig=AQAAAJpTMCW9TlfoMsU9T31eDclwTClE3c-sNeq-K8AnjALcZe_Kxd9Lx_uvNjJFHkVALwVkaGzpRLm0PsJcuaXVKgOTC5nytbmSGd1SIqfJtQ9jrKvLSqq6MdXVktsQoEHUyiKkbCb-IBBZZjMjjc6gFLsF8riaAOAjyjL-kIY3iA_m\" target=\"_blank\">number of factors<\/a>&nbsp;helping reach their amazing age, including chocolate consumption, treating their skin with olive oil, smoking cigarettes (!), and eating sushi.<\/p>\n<p>But genetics of supercentplays a role, too. According to Boston University&rsquo;s <a href=\"https:\/\/www.bumc.bu.edu\/centenarian\/\" target=\"_blank\">New England Centenarian Study<\/a>, genetics plays a role in those who live into their 90s and beyond.<\/p>\n<p>&ldquo;This genetic influence probably involves many genetic variants with individually modest effects, but as a group, they have a strong effect,&rdquo; the study&rsquo;s website notes. &ldquo;But for some rare exceptions, centenarians have just as many disease-associated genetic variants as the average population. Thus, <strong>their genetic advantage is likely due to variants that slow aging and decrease risk for aging-related diseases such as heart disease, stroke, cancer, diabetes and Alzheimer&rsquo;s Disease.&rdquo;<\/strong><\/p>\n<p>The ongoing study found 281 genetic markers that are 61% accurate in predicting who will make it to 100, and that are 85% accurate in predicting who lives on to 105. Those markers point to 130 genes which play roles in conditions like cancer, high blood pressure, and heart disease.<\/p>\n<p>As described by <a href=\"https:\/\/nyti.ms\/2hw8W32\" target=\"_blank\">The New York Times<\/a>, pinning down what that genetic variant is has proven to be hugely difficult&mdash;actually, even finding people that old whose age can be verified is pretty tough, thanks to spotty record-keeping.<\/p>\n<p>A <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC4229186\/\" target=\"_blank\">2014 study<\/a>&nbsp;of supercentenarians went looking for a long life gene&mdash;and may have found one.<\/p>\n<p>In that study, researchers performed <a href=\"\/pages\/technology\/next-generation-sequencing\/dna-sequencing\/targeted-sequencing\/exome-sequencing\">whole exome sequencing<\/a>&nbsp;of 17 supercentenarians, performed an annotation, filtered out common variants, and collapsed all variants into genes, from which one stood out: <em><a href=\"https:\/\/www.genecards.org\/cgi-bin\/carddisp.pl?gene=TSHZ3\" target=\"_blank\"><em>TSHZ3<\/em><\/a>. <\/em>They also found one person with a known pathogenic allele in gene <a href=\"https:\/\/www.genecards.org\/cgi-bin\/carddisp.pl?gene=DSC2\" target=\"_blank\"><em>DSC2<\/em><\/a>, which is associated with the potentially fatal condition known as arrhythmogenic right ventricular cardiomyopathy.<\/p>\n<p>&ldquo;The presence of this mutation in the DNA sequence of a young person today should be reported to him\/her and their families with caution, as it may or may not result in arrhythmogenic right ventricular cardiomyopathy,&rdquo; researchers wrote. &ldquo;Generally, variants that are annotated as pathogenic are of unknown penetrance.&rdquo;<\/p>\n<p>A <a href=\"https:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0002421\" target=\"_blank\">2008 study<\/a>&nbsp;of extreme longevity in Japan, meanwhile, identified another variable: <strong>mitochondrial DNA haplogroup <a href=\"https:\/\/en.wikipedia.org\/wiki\/Haplogroup_D_(mtDNA)\" target=\"_blank\">D4a<\/a><\/strong>&mdash;an Asian haplogroup contemporarily found in central and northeast Asia. Researchers then analyzed the data to see if the longevity was due to population structure or a property of a mutation.<\/p>\n<p>&ldquo;The conclusion is that the correlations are entirely due to population structure (phylogenetic tree). We find no signal for a functional <a href=\"\/pages\/products\/next-generation-sequencing\/workflow\/xgen-ngs-hybridization-capture\/pre-designed-hyb-cap-panels\/human-mtdna-hyb-panel\">mtDNA<\/a>&nbsp;SNP correlated with longevity,&rdquo; researchers wrote. &ldquo;The fact that the correlations are from the population structure suggests that hitch-hiking on autosomal events is a possible explanation for the observed correlations.&rdquo;<\/p>\n<h2>What is the maximum age can a human live?<\/h2>\n<p> According to an <a href=\"https:\/\/link.springer.com\/article\/10.1007\/s10522-008-9156-4\" target=\"_blank\">article<\/a>&nbsp;in <em>Biogerontology<\/em>, the <strong>theoretical maximum age a person can currently be is 126.&nbsp; <\/strong>Besides DNA, longevity in humans is often tied to diets, and in particular the so-called <a href=\"https:\/\/www.netflix.com\/title\/80230601\" target=\"_blank\">Blue Zone diet<\/a>, a set of common foods found in the areas of the world where people live the longest, including long-life hotspots like the Italian island of Sardinia.<\/p>\n<p>But evolving <a href=\"\/pages\/technology\/crispr\">CRISPR technologies<\/a>&nbsp;could upend that. Recently, <a href=\"https:\/\/www.sciencedaily.com\/releases\/2019\/02\/190219111747.htm\" target=\"_blank\">scientists found<\/a>&nbsp;that Cas9 therapy can slow aging, improve health, and extend lifespans in mice. That therapy involved the injection of an adeno-associated virus with two synthetic guide RNAs and a reporter gene; the <a href=\"\/pages\/products\/crispr-genome-editing\/custom-guide-rna\">guide RNA<\/a>&nbsp;led the Cas9 protein to a specific DNA location where it disrupted two proteins associated with early-onset dementia, lamin A and progerin.<\/p>\n<p>Aging itself is a risk factor for debilitating conditions such as cancer and Alzheimer&rsquo;s, <a href=\"http:\/\/dx.doi.org\/10.1038\/s41591-019-0343-4\" target=\"_blank\">noted a 2019 study<\/a>, meaning that longevity alone is not a key to better living. What is needed is an actual anti-aging process, which CRISPR may be able to deliver.<\/p>\n<p>In that study, published in <em>Nature Medicine<\/em><strong>, Cas9 gene editing was found to suppress the accelerated aging <\/strong>usually seen in mice with Hutchinson-Gilford progeria syndrome&mdash;a rare disorder which also affects humans. An adeno-associated virus with two <a href=\"\/pages\/education\/decoded\/crispr-genome-editing\" target=\"_blank\">synthetic guide RNAs<\/a>&nbsp;and a reporter gene was injected into study mice. Two months later, the mice were healthier, stronger, and more active, and overall, their lifespan increased about 25%.<\/p>\n<p>While CRISPR is by no means yet a ticket to eternal youth, it&rsquo;s being tested in the fight against <a href=\"https:\/\/www.nature.com\/articles\/nature17946\">Alzheimer&rsquo;s<\/a>&nbsp;and <a href=\"\/pages\/community\/blog\/post\/using-ngs-to-trace-the-origins-of-malaria-was-it-asia-or-africa\" target=\"_blank\">malaria<\/a>, with more possibilities on the horizon.
