IDT releases new mutagenesis application guide

CORALVILLE, IA – Nov 1 2011. Integrated DNA Technologies (IDT), the world leader in oligonucleotide synthesis, has recently released its free to download mutagenesis applications guide. Providing an overview of in vitro mutagenesis, along with in-depth protocols and troubleshooting advice, the guide makes it easy to carry out efficient and effective experiments. Supplying a wealth of useful details, the guide describes traditional mutagenesis approaches, including site directed mutagenesis by polymerase chain reaction (PCR), primer extension, inverse PCR and cassette-based methods. The guide also covers random mutagenesis, a technique which is often used to optimize protein expression or function by creating unpredictable alterations.

One approach to mutagenesis detailed within the guide is the use of IDT’s Ultramer™ oligos, which simplifies the addition of large changes into a target sequence, providing greater experimental flexibility. This is achieved since Ultramers are the highest fidelity long nucleotides currently available, with lengths varying from 60-200 bases. As such, they enable primers to be extended into regions that have traditionally been difficult to target, with more optimal sequence composition than was previously attainable.
