What purification do I need for my NGS library amplification primers?

Next generation sequencing (NGS) library amplification primers that are simply desalted show no noticeable performance consequence over primers that have undergone HPLC purification, suggesting that the latter purification is typically unnecessary. Library amplification primers of 20–25 bases from IDT already have an 83–85% molar purity based on the industry leading coupling efficiency IDT achieves with our oligonucleotide synthesis technology. See the DECODED article, Getting Enough Full-Length Oligo?, for data and a longer discussion on the importance of coupling efficiency. Also, feel free to contact our NGS specialists at xgen@idtdna.com to discuss further.
