When should I choose PAGE vs HPLC purification?

IDT recommends PAGE purification for long unmodified oligos and oligos used in cloning, gel shift, and mutagenesis protocols.

Due to their hydrophobicity, most oligos with modifications are better separated by HPLC. Therefore, IDT requires HPLC purification of oligos with specific modifications. HPLC routinely provides 85% purity, while PAGE provides 90% purity.

The final yield of a PAGE purified oligo, however, will be less than an HPLC purified oligo.
