Where should I place phosphorothioate bonds to increase stability of my oligo?
Including 3–5 nucleotides with phosphorothioate bonds at the 3' and 5' ends of your oligo can inhibit exonuclease degradation. Including phosphorothioate bonds throughout the entire oligo will help reduce attack by endonucleases as well.
