What is the workflow for xGen™ DNA Library Prep Kit MC library construction?

The xGen DNA Library Prep Kit MC protocol contains three enzymatic incubations and two bead-purification steps, thereby minimizing sample handling and overall library preparation time to under two hours.

  • The first incubation consists of end repair, polishing, and dA-tailing, all performed in a single End Prep reaction.
  • The second incubation performs ligation of either full-length indexed Y adapters, or stubby Y adapters and a bead-purification step.
  • The third incubation is PCR amplification which completes the indexed adapters (when ligating stubby Y adapters) for increased library yield, followed by a second bead-based clean-up.
