Engineering an improved inhibitor of 53BP1 to enhance HDR efficiency

Precise genome editing can be achieved using a targeted nuclease that introduces a dsDNA break which can then be repaired by endogenous repair pathways. A modified variant of ubiquitin was shown to bias DNA repair machinery toward a pathway that allows for incorporation of an exogenous DNA template. Here we mutagenized and isolated ubiquitin variants that allow for highly efficient gene replacement following CRISPR cleavage in livings cells.
